Glucosylation of O-Antigen in Salmonella Carrying El5 and Es4 Phages*

چکیده
Enzymatic reactions involved in the O-34 antigen formation of group E3 Salmonella carrying phages cl3 and ea4 was studied in vitro by the use of cells defective in UDP-glucose4-epimerase activity. Incorporation of glucose from UDPglucose into O-34 antigen required polymerization of mannosylrhamnosylgalactose repeating units. A single trisaccharide unit was not an acceptor for O-34 glucose. A preformed trisaccharide repeating polymer generated under restricted conditions was able to accept glucosyl residues. The product of glucose incorporation was characterized and found to be a haptenic 0-polysaccharide. A glucosyl-lipid intermediate was formed, which acted as a glucosyl donor to the polymeric trisaccharide units. Glucose incorporated into the intermediate in the first step reaction was transferred to the mannosylrhamnosylgalactose polymer generated in the second step. The following two enzymatic steps were formulated.

A group of Escherichia coli and Salmonella enterica O antigens sharing a common backbone structure.
The O-antigen moiety of the LPS is one of the most variable cell surface components of the Gram-negative bacterial outer membrane. Variation is due to the presence of different sugars and sugar linkages. Here, it is reported that a group of Escherichia coli O serogroups (O17, O44, O73, O77 and O106), and the Salmonella enterica serogroup O : 6,14 (H), share a common four-sugar backbone O-subuni...
متن کاملA conserved acetyl esterase domain targets diverse bacteriophages to the Vi capsular receptor of Salmonella enterica serovar Typhi.
A number of bacteriophages have been identified that target the Vi capsular antigen of Salmonella enterica serovar Typhi. Here we show that these Vi phages represent a remarkably diverse set of phages belonging to three phage families, including Podoviridae and Myoviridae. Genome analysis facilitated the further classification of these phages and highlighted aspects of their independent evoluti...
متن کاملReceptor Diversity and Host Interaction of Bacteriophages Infecting Salmonella enterica Serovar Typhimurium
BACKGROUND Salmonella enterica subspecies enterica serovar Typhimurium is a gram-negative pathogen causing salmonellosis. Salmonella Typhimurium-targeting bacteriophages have been proposed as an alternative biocontrol agent to antibiotics. To further understand infection and interaction mechanisms between the host strains and the bacteriophages, the receptor diversity of these phages needs to b...
متن کاملEpigenetic Control of Salmonella enterica O-Antigen Chain Length: A Tradeoff between Virulence and Bacteriophage Resistance
The Salmonella enterica opvAB operon is a horizontally-acquired locus that undergoes phase variation under Dam methylation control. The OpvA and OpvB proteins form intertwining ribbons in the inner membrane. Synthesis of OpvA and OpvB alters lipopolysaccharide O-antigen chain length and confers resistance to bacteriophages 9NA (Siphoviridae), Det7 (Myoviridae), and P22 (Podoviridae). These phag...
متن کاملA virulent isolate of Salmonella enteritidis produces a Salmonella typhi-like lipopolysaccharide.
The lipopolysaccharide (LPS) of Salmonella enteritidis has been implicated as a virulence factor of this organism. Therefore, the LPS from a stable virulent isolate, SE6-E21, was compared with that from an avirulent isolate, SE6-E5. The LPSs were extracted, and the high-molecular-weight (HMW) LPS was separated from the low-molecular-weight (LMW) LPS for both isolates. Both the HMW and LMW LPSs ...
